First, I'd just play around - you can learn a lot by experimenting. When that isn't enough, turn to the built-in Help files. When the "?" tab in the lower right cornor of the NXT Software window is selected, clicking on a block will bring up a very brief description of the block, with a link to more information in the form of a detailed help page. There are help pages for every block, as well as subjects like data wires, sequence beams, and other aspects of the programming language. You can also access this built-in resource by using the "Help" menu.

When you need to learn beyond that, it's sometimes helpful to look at other peoples ideas and code, such as the examples on NXTLOG or elsewhere - there's a big LEGO MINDSTORMS community you can tap for help. Finally, when you've exhausted all those resources, there are some books out there that provide a detailed description of some aspects of the programming language. One is Jim Kelly's book, LEGO MINDSTORMS NXT-G Programming Guide. It's easy to understand and covers pretty much everything.
